Verlie Lines grew up in rural Queensland, attending correspondence school in her early years and then finishing her formal education at Parramatta High School at the early age of 16 years. She won an Exhibition to the University of Sydney, where she studied medicine, graduating in 1950.

Lines gained her Diploma of Anaesthesia from the University of Melbourne in 1954, and followed that with admission to the fellowship of the Faculty of Anaesthetists in 1956. She undertook her training in Melbourne, Tasmania and the United Kingdom.

Returning to Sydney, she took up a position as staff anaesthetist with the Royal Alexandra Hospital for Children in 1955, and held that position for the following 30 years.
