In 1947, Paul Danby Kempthorne graduated from medicine at the University of Otago. In 1955 he gained a Diploma of Anaesthesia in London and was elected to fellowship of the Faculty of Anaesthetists of the Royal College of Surgeons that same year.

Prior to going to England, Kempthorne was an anaesthetic registrar at Wellington Hospital. Upon his return, he became visiting anaesthetist there until 1955. Kempthorne was appointed director of anaesthesia at Waikato Hospital in 1955, a position he held until 1986.
